Abstract
The plasma membrane, one of the most fundamental parts of the cell, acts as an efficient barrier between the cell's interior and its environment, regulating the movement of substances in and out. This protective function also prevents therapeutic agents from easily penetrating the membrane, making drug delivery a challenge. In the context of the matter that enters the cell, special attention was given to cell-penetrating peptides (CPPs), particularly those with a cationic-hydrophobic character, that deliver cargo into cells without disrupting their structure. Their adsorption on the membrane represents the first step of their internalization mechanism and is strongly influenced by the physicochemical properties of membrane lipids, particularly the charge of lipid headgroups. This chapter introduces temperature-dependent UV-Vis spectroscopy as a simple and sensitive approach for characterizing charge-regulated interactions between CPPs and lipid bilayers composed of a single lipid, either phosphatidylcholine (PC), phosphatidylglycerol (PG), or phosphatidylserine (PS). The thermotropic features of lipid vesicles in the absence and presence of selected cationic-hydrophobic peptides are analyzed and compared with conventional differential scanning calorimetry (DSC) measurements. Particular emphasis is placed on the ability of UV-Vis spectroscopy to detect subtle peptide-induced alterations in membrane organization, lipid packing, and phase transitions that arise from differences in lipid headgroup charge.
