Abstract

In this study, we showcased the effectiveness of UV-Vis spectroscopy for characterizing the thermotropic properties of mixed lipid bilayers composed of phosphatidylcholine (PC), phosphatidylglycerol (PG), and either stearic acid (SA) or linoleic acid (LA), in the presence of the antimicrobial peptide anoplin (ANP). We measured the phase transition temperatures of these lipid bilayers using differential scanning calorimetry (DSC) as the standard method, and temperature-dependent UV-Vis spectroscopy. The advantage of UV-Vis spectroscopy stems from its ability to detect events that DSC does not register. Additionally, it demonstrated greater sensitivity in identifying differences in lipid bilayer phase transition temperatures depending on whether SA or LA was present, as well as in assessing the impact of ANP on membrane fluidity. Supplemented by measurements of lipid bilayer sizes, the results of this study highlight the potential of UV-Vis spectroscopy in identifying structural changes in lipid bilayers associated with membrane permeabilization.
