Abstract

Melanoma cell line MDA-MB-435S forms integrin αVβ5 mediated focal (FA) and reticular adhesions (RA). Knockdown of integrin αV increases the sensitivity of MDA-MB-435S cells to the microtubule-targeting antitumor drug paclitaxel and reduces migration. In this work, we showed that the mechanism of these effects is mediated by the binding of talin 2 from FA and KANK2 from the cortical microtubule stabilizing complex. We identified talin 2 as a potential target in tumor therapy. We analyzed RA in MDA-MB-435S cells and discovered KANK2 as a new component of RA.
